R)-(+)-Lipoic Acid Powder, also known as dextro-lipoic acid, is a white or yellowish, odorless, powdery or crystalline substance commonly used in vitamin drugs for the treatment of chronic hepatitis, psoriasis, and erythematous lupus.

R)-(+)-Lipoic acid CAS 1200-22-2 is a yellow crystalline substance. Lipoic acid is a biologically active natural product.In 1951 Raed et al. reported the isolation of a crystalline cofactor for growth-promoting enzymes from treated insoluble liver tissue residue.The compound was named Lipoic acid because of its highly fat-soluble and acidic nature (pka = 4.7).

The two enantiomers of Lipoic acid show different biological activities. Among them, the R-type enantiomeric isomer was more effective than the S-type, which may be due to the fact that during the metabolism of lipoic acid, a large amount of R-type lipoic acid could enter the cell and mitochondria through the cell membrane and mitochondrial membrane to be reduced to dihydrolipoic acid while only a small amount of the S-type entered into the cell and was reduced. Dihydrolipoic acid has a stronger antioxidant capacity than lipoic acid, and the regeneration of endogenous antioxidants and the repair of oxidative damage can only be realized through the form of dihydrolipoic acid.

Production methodprocess of R)-(+)-Lipoic acid Powder

• Taking ethyl 6,8-dichlorooctanoate as the starting material, through thiosulfuration, cyclization and hydrolysis, the racemate α-lipoic acid is obtained (U.S.2792406,1957; U.S.3223712,1965), and then R-lipoic acid can be obtained by splitting with a splitting agent, and the yield of this step is only 45%. Moreover, S-lipoic acid is not easy to be converted into R.S-type or R-type, which wastes half of the resources and makes R)-(+)-Lipoic acid Powder
• The cost of R)-(+)-Lipoic acid Powder is expensive. Methyl 6,8-dihydroxyoctanoate or methyl 6-hydroxy-8-chlorooctanoate was used as the starting material to make methanesulfonate, and then stereoselectively converted into R-lipoic acid (U.S. 5869713, G. Bringmann, J. Paust Z. Naturforschung 54b,661-665, 1999), but the process was more complicated, and was not easy to obtain R)-(+)-Lipoic acid CAS 1200-22-2.
• Hydrolysis of ethyl racemic 6, 8-dichlorooctanoate to (±)dichlorooctanoic acid (DCA), salting with (-)ephedrine, splitting, rethiolation, cyclization (Acker et al., J. Am. Chem. Soc., 76,6483,1954), which is basically cost-saving, but still leaves about 50% of S-(-)-6, 8-dichlorooctanoic acid unutilized at a higher cost。
